二、核心能力與技術基礎
1. 糖科學(Glycosciences)技術平臺
GlycoDiag的核心技術平臺圍繞凝集素與糖鏈之間的相互作用展開,旨在識別和比較特定的糖基化特征。其GLYcoPROFILE技術采用標準微孔板形式,無需昂貴的專用設備,適用于高通量篩選,且樣品無需純化即可進行分析,適合工藝優化與過程控制。
基于該技術,公司開發了LEctPROFILE系列試劑盒,用于快速評估粗提或純化糖復合物與凝集素的相互作用,檢測時間通常在3小時以內。該系列涵蓋多個靶向產品:用于檢測末端半乳糖(Galili表位)的Galili LEctPROFILE® kit、用于研究GalNAc糖鏈結構的ASGPR LEctPROFILE® kit、用于甘露糖化糖鏈結構檢測的DC-SIGN & Langerin LEctPROFILE® kit及FimH LEctPROFILE® kit、用于β-半乳糖苷結構研究的Galectin 3 LEctPROFILE® kit,以及用于細胞表面糖基化分析的Cells LEctPROFILE® kit等。此外,Glycoprotein LEctPROFILE® kit支持在24種凝集素上對糖蛋白進行糖基化水平的系統分析,可用于糖蛋白批次間一致性監測。

2. 微生物群(Microbiota)研究體系
在微生物群領域,GlycoDiag建立了基于野生型菌株的體外共培養模型,這些菌株分離自健康志愿者的特定部位(如面部),相較于標準實驗室菌株更能反映自然行為。共培養模型通常由4至5種代表特定皮膚微環境的細菌和/或酵母組成,在同一條件下共同生長。
目前覆蓋的皮膚微生物生態系統包括:干性、濕性、油性皮膚、頭皮、面部、口腔等。該模型已用于評估化妝品成分對微生物群的影響,結果可區分為殺菌、益生元或維持菌群平衡(“微生物群友好")等作用。其部分研究成果已通過同行評審發表。
三、解決實驗中的哪些問題?
1. 糖基化分析與質控問題:在生物治療藥物研發中,早期檢測非預期的糖基化是質控的重要環節。LEctPROFILE試劑盒可在工藝開發階段提供糖基化特征的快速反饋,無需復雜的樣品前處理即可完成。
2. 糖蛋白表征與批次一致性:重組糖蛋白生產過程中,批次間的糖型差異可能影響療效與安全性。Glycoprotein LEctPROFILE® kit能夠在24種凝集素層面提供糖基化模式的可視化比對,輔助監測工藝穩定性。
3. 微生物群影響評估的模型問題:化妝品及營養品行業在驗證原料對皮膚或腸道微生物群的影響時,常受限于缺乏生理相關的體外模型。GlycoDiag基于野生型菌株的共培養模型模擬了特定微環境的溫度、pH及菌群比例,為評估產品是否“微生物群友好"提供了實驗依據。
4. 候選分子篩選:LEctPROFILE試劑盒的競爭模式可用于篩選特定凝集素的拮抗劑,評估候選分子的半抑制濃度(IC50)或親和力比較,適用于藥物發現階段的早期篩選。
四、常見問題與解答
問:試劑盒是否只能用于純化樣品?
答:不是。LEctPROFILE試劑盒可直接用于粗提樣品或未純化的混合物分析,無需對樣品進行預先純化,這簡化了工藝優化過程中的樣品處理流程。
問:使用這些產品是否需要專門的糖生物學知識或設備?
答:GLYcoPROFILE技術在設計上考慮了非專業人士的使用需求,基于標準微孔板平臺,可利用常規實驗室的酶標儀進行讀數,無需購置專用設備。試劑盒提供標準化的操作流程和已驗證的試劑,便于在自有實驗室中轉移實施。
問:共培養模型使用的是標準菌株還是更具代表性的菌株?
答:GlycoDiag優先使用從健康志愿者特定部位(如面部、頭皮)分離的野生型菌株,而非僅依賴標準實驗室菌株。這些野生型菌株保留了與原始微環境相關的自然行為特征,在模擬生理條件下可能更具代表性。
問:這些產品和服務是否適用于臨床診斷?
答:不適用。LEctPROFILE試劑盒及相關產品明確標示為“僅限研究使用"(For research use only),不適用于臨床診斷目的。
